Improvement of Detection Sensitivity of Listeria monocytogenes Using Crude Pediocin PA-1 by Polymerase Chain Reaction (PCR)

Abstract

|
Listeria monocytogenes is a gram-positive bacteria with a lysis resistant cell wall composed of rigid peptidoglycan. In general, L. monocytogenes cna be detected at the 10^(£³) cell level usng a species-specific primer set from artificially contaminated food samples. When treated with 160 AU of crude pediocin PA-1, which is an antilisterial bacteriocin produced by Pediococcus acidilactici K10, the detection limit of L. monocytogenes was reduced to 10^(£±) and 10^(£²) cells from artificially contaminated raw pork and whole milk, respectively. By using crude pediocin PA-1, the cell membrane of the pathogen was effectively disrupted to release DNA, indicating an increase in susceptibility to PCR detection. The total assay time for the food-borne L. monocytogenes treated with the crude pediocin PA-1 was £¼4 hr without enrichment at a high sensitivity.
